Optimization of Preparation Processing of Deer Protein and Peptide and Its Proliferation Activity on TM3 Cells

Abstract
Taking the sika deer tail as research object,ultrasonic water extraction of protein technology and water enzymatic extraction of peptides were optimized by orthogonal test. The proliferation activity of TM3 cells was compared between the two optimally obtained proteins. The results showed that the optimal ultrasonic water extraction of protein process were as follows:The ratio of material to liquid was 1:5 g/mL,the ultrasonic power was 200 W,the extraction time was 1 h,and the extraction temperature was 40 ℃,the content of protein was 60.72%±0.56% during the condition. The best protease of water enzymatic extraction of peptides was trypsin. The optimal conditions for enzymatic hydrolysis were pH8,enzyme amount addition 1500 U/g,enzymatic hydrolysis for 4 h at 50 ℃,and degree of hydrolysis was 22.85%±0.35% during the condition. The enzymatic extraction of deer tail polypeptide on the proliferation of TM3 was superior to that of ultrasonic water extracting deer tail protein.
