Purification of Flavonoids from Red Raspberry Seed by Macroporous Resin and the Ability of Bile Salt-binding in Vitro

Abstract
The purification conditions of red raspberry seed flavonoids by AB-8 macroporous resin were investigated and its ability of binding bile salts was evaluated. The optimal purification conditions of AB-8 macroporous resin was as follows:Sample concentration was 10 mg/mL,sample volume was 100 mL,the sample flow rate was 1.5 mL/min and sample pH was 4.5,the volume fraction of the ethanol was 60%,the volume of elution was 150 mL,and the flow rate of elution was 1.5 mL/min. The purity of red raspberry seed flavonoids was 48.19% and which was 23.12% higher than those unpurified. And four kinds of extraction methods for the three kinds of bile salt binding ability was in the order:purified by AB-8,ethanol extracted,ultrasonic extracted and enzyme extracted flavonoids. Moreover,hypolipidemic experiments showed that purified flavonoids by AB-8 had strong combining capabilities for sodium taurocholate,sodium glycocholate,sodium cholate with combination rate of 79.35%,74.88% and 74.56%. The results showed that the red raspberry seeds flavonoids had ability to bind to bile salts,among which AB-8 had the strongest binding ability.
