Determination of Biochemical Components of “Dashancha” from Fengqing of Yunnan

Abstract
The plantation area of tea plant cultivar "Dashancha" was increasing sharply in recent years. However, the germplasm was not evaluated. In this work, sun-dried tea leaves were produced by the one bud and two leaves of "Dashancha" and tea plant cultivars of Yunkang-10#, etc. The biochemical components were detected and compared. Contents of water extraction, tea polyphenols, free amino acids and polysaccharide in "Dashancha" were 48.77%±0.26%, 36.01%±0.92%, 1.66%±0.09% and 7.76%±0.99%, respectively, by ultraviolet spectrophotometric method. High performance liquid chromatography (HPLC) analysis showed the sum of contents of catechins was (154.49±31.54) mg/g, and level of caffeine was (17.42±1.44) mg/g. 44 compounds were detected in methanol extraction of Dashancha by LC-MS/MS. The determination of catechins showed that three peak areas in Dashancha were high than that in same quality of Yunkang-10# tea. Two of them were identified as 3-Galloylquinic acid and Corilagin. The biochemical components in "Dashancha" were demonstrated preliminary. And provide a base for further germplasm evaluation.
