Abstract
Objective: To explore the optimum extraction process of polysaccharide from the ink of the Sthenoteuthis oualaniensis in South China Sea,response surface methodology( RSM) was employed to optimize extraction process of polysaccharide from the ink of the Sthenoteuthis oualaniensis. Methods: Experiment factors and levels were first selected on the ground of single factor experiment.Along with the center combination experiment design principle,three factors and three levels of response surface analysis were adopted to obtain a multiple quadratic linear regression equation and analyzed the interaction among factors chosen Y( polysaccharide content × total sugar quality,100 g ink extraction product of quality of crude polysaccharide and total sugar content%) as the response value. Results: The optimum extraction conditions of polysaccharide from the ink of the Sthenoteuthis oualaniensis in South China Sea were concluded as follows: ratio of solid to liquid 1∶ 2,water extract p H5.5,enzyme concentration 1.00%.Under this condition,polysaccharide content and crude polysaccharide extraction yield value of polysaccharide from the ink of the Sthenoteuthis oualaniensis was 9.8% and 0.91% respectively.Conclusion: The RSM was employed to optimize extraction process of polysaccharide from the ink,which had feasibility and good effect.
