Study on ultrasonic- assisted solvent extraction and purification of pepsin- soluble collagen from grass carp skin

  • Ultrasonic- assisted solvent extraction and purification pepsin- soluble collagen from grass carp skin was investigated in this paper.Ultrasonic rate,ultrasonic time and enzyme extraction time were chosen as influencing factors.The extraction rate of collagen was selected as response value.The mathematical model was established by Box- Behnken central composite design.The analysis method was response surface methodology( RSM).The final optimum conditions were: ultrasonic rate 310 W,ultrasonic time 10 min and enzyme extraction time 29 h,extraction rate of collagen was 68.67%,it's nearly to the model 68.8%,relative tolerance was 0.189%.SDS- PAGE and FTIR showed that ultrasonic- assisted extraction hadno obvious effect on collagen structure.
