Identification and detection of flavonoids in bee pollen

  • To establish a quick,accurate and sensitive approach to analyse flavonoid contents using 24 kinds of bee pollen as material,a method called ultra- high pressure liquid chromatograph with tandem triple level 4 pole mass spectrometry had been set up in this research,aimed to detect 8 types of flavone.After crushed the wall of samples by temperature,differential method stepped by the acid hydrolysis through ethanol extraction and finally diluted with methanol solution,the targets aimed to detect were seperated by reversed- phase chromatography,measured by mass spectrometry and quantitated by external standard method. Statistics indicated that this method had a good linear with R2> 0.999,its RSD was less than 6.6 percent and more than 3.2 percent. The LOD was between0.15 and 3.0 mg / kg along with a hight recovery rate ranged from 91.2% to 100.2%. Depended on its easier operation,reliable result with lower matrix interference,this method could be applied to other flavonoids contents in bee pollen such as quercetin,kaempferol,rutin and so on.Last but not the least,it could also provide experimental bases and research methodology to identify the plant origin for bee pollen,distinguish authenticity further to build revelant fingerprint.
