石榴皮单宁的提取、纯化及抑菌作用的研究

Study on extraction, purification and antimicrobial activity of pomegranate peel tannins

  • 摘要: 石榴皮中含有丰富的单宁类物质,具有良好的抑菌活性。以丙酮为提取溶剂,采用超声波辅助法对石榴皮单宁进行提取,并用Amberlite XAD-16大孔吸附树脂纯化,经高效液相色谱对其组分进行分析后,采用琼脂稀释法做最小抑菌浓度实验,结果表明:石榴皮提取物的主要成分为安石榴苷,纯化后质量浓度由12.3%提高到64.2%;石榴皮粗提物对金黄色葡萄球菌、单核细胞增生李斯特氏菌、沙门氏菌和大肠杆菌的最小抑菌浓度分别为0.312、10、0.625mg/mL和大于10mg/mL,纯化物对四种菌的最小抑菌浓度分别为0.156、5、0.625、10mg/mL。 
    Abstract: Pomegranate peel is rich in tannins which have a good antibacterial activity. Pomegranate peel tannins were extracted by acetone in ultrasonic-assisted extraction method. After that they were purified by Amberlite XAD-16 macroporous resin. Then high performance liquid chromatography (HPLC) analysis was taken to determine the components and agar dilution method was adopted to test the minimal inhibitory concentration (MIC) . The results indicated that the concentration of punicalagin, the main component of pomegranate peel extract, increased from 12.3% to 64.2% after purification. The MICs of pomegranate peel crude extract on Staphylococcus aureus, Listeria monocytogenes, Salmonella and E.coli were 0.312, 10, 0.625mg/mL and larger than 10mg/mL, while the MICs of purified extract for these four kinds of bacteria were 0.156, 5, 0.625 and 10mg/mL respectively.
